The thesis aimed at investigating the predictive values of the tests used in the diagnostic work up of penicillin allergy. Patients with penicillin-induced positive skin tests and/or positive specific IgE to penicillin were challenged with penicillin to investigate the clinical relevance of a positive test. Reproducibility of the diagnostic tests was studied by repeated skin testing and IgE measurement. The thesis also dealt with mechanistic aspects of the diagnostic tests: Skin microdialysis was done to study histamine release in a penicillin-induced positive skin test, and the functionality of specific IgE to penicillin was elucidated by applying the Prausnitz-Küstner technique.
